Pigpen is an endothelial protein of the Ewing's sarcoma family that parallels the transition from quiescent to angiogenic phenotypes in vitro. Using YPEN-1 endothelial cells and N9 micorglial cells, respectively, in a selection and counter-selection strategy in SELEX, a DNA aptamer named III.1 was isolated that preferentially bound to YPEN-1 cells.68 The III. 1 was also shown to selectively bind to the microvessels of experimental rat glioblastoma. The isolation and characterization of the III.1 target identified pigpen as the target antigen. The use of III.1 aptamer for targeting the microvasculature of tumors is a potentially powerful means of delivering drugs to the site of the cancer.
